Tier 1: sleep — the best-evidenced consumer benefit

This is glycine's strongest, most direct evidence. In the best PubMed-indexed human trial, volunteers whose sleep had been experimentally restricted took 3 g of glycine before bed and showed reduced next-day fatigue and better subjective daytime performance compared to placebo (Bannai 2012, PMID: 22529837). That's a real, modest effect on how you feel the next day — not a demonstrated cure for insomnia, and the more sweeping "falls asleep faster, better sleep quality" claims you'll see repeated online trace back to an older pilot study that isn't indexed in PubMed, so it doesn't meet this site's citation bar.

There's a plausible mechanism behind the effect, too: glycine appears to lower core body temperature by acting on NMDA receptors in the brain's suprachiasmatic nucleus, the region that governs your sleep-wake clock — a drop in core temperature is part of how the body signals it's time to sleep (Kawai 2015, PMID: 25533534). That mechanistic study was done in animals, not humans, so treat it as a reasonable explanation for the human sleep finding above, not separate proof of its own. Tier 2: glutathione precursor, and the GlyNAC combination in older adults

Glycine is one of the three amino acids your body needs to build glutathione, its main intracellular antioxidant, and a review of the biochemistry argues that ordinary dietary glycine intake may not always be enough to keep glutathione synthesis running at full capacity — tissue glycine levels tend to sit below the concentration the synthesizing enzyme needs for maximum output (McCarty 2018, PMID: 29559876). That paper is a literature review making a mechanistic case, not a clinical trial, and its own conclusion is that "clinical research is warranted" — a plausible rationale, not proof that a glycine pill measurably raises your glutathione.

The more concrete evidence comes from combining glycine with N-acetylcysteine (NAC), a pairing known as GlyNAC, since both are needed for glutathione synthesis. A placebo-controlled randomized trial gave older adults either GlyNAC or an inactive placebo for 16 weeks (12 people per arm, with 12 young adults studied as a reference group) and found the GlyNAC group — not the placebo group — showed improved glutathione deficiency, reduced oxidative stress, better mitochondrial function, lower inflammation, and gains in physical function measures like gait speed and grip strength (Kumar 2023, PMID: 35975308). That's a genuinely promising, well-designed result. Two caveats matter: it's a small trial (12 per arm) that needs replication at larger scale, and it tested the glycine-plus-NAC combination, so it doesn't tell you what glycine does on its own. Tier 2: collagen synthesis — a real building-block role

Glycine makes up roughly a third of the collagen molecule, and a lab study using cultured bovine cartilage cells found that raising glycine concentrations increased type II collagen synthesis by 60 to 75%, more than the increases produced by two other amino acids collagen needs (proline and lysine) (de Paz-Lugo 2018, PMID: 30006659). That's a real finding, but it's an in vitro cell-culture study, not a human trial — it shows glycine can drive collagen production in a dish, not that taking glycine supplements measurably improves anyone's skin, joints, or cartilage. Worth noting for transparency: the senior author's research institute has applied for a patent related to using glycine for osteoarthritis, a disclosed conflict of interest that doesn't invalidate the lab result but is a reason to wait for independent, human confirmation before treating this as settled. Tier 3: metabolic and blood-sugar claims — the thinnest evidence

This is where to be most skeptical. It's well-replicated that people with insulin resistance and type 2 diabetes tend to have lower blood glycine levels, which makes glycine a useful marker of metabolic health (Adeva-Andany 2018, PMID: 29094215).
