The dose depends on which trial's evidence you're chasing — and retail usually overshoots it
Spermidine's dosage story runs the opposite direction from a lot of supplements: instead of retail underdosing the trial amount, retail is frequently ABOVE any efficacy-tested dose. The two trials that measured a cognitive outcome used tiny fractions of a milligram compared to what's on the shelf. That earlier pilot, Wirth 2018 (n=30, 3 months), tested 1.2mg/day and found improved memory discrimination (Cohen's d=.77) — a real but tiny, unreplicated-at-scale result. Read those two together, chronologically: a promising pilot at 1.2mg, an adequately powered confirmatory trial at 0.9mg that didn't confirm it on its primary outcome. Separately, Keohane 2024 (n=37 men, 28 days) pushed the dose to 40mg/day — roughly 30-40× the trial-efficacy doses — but that trial measured safety and pharmacokinetics only, no cognitive or clinical endpoint, and even at that much higher dose, circulating spermidine levels barely moved. That's an open bioavailability question, not evidence that 40mg works better than 0.9–1.2mg — no trial has ever tested that comparison for efficacy. Meanwhile, retail products that disclose an actual spermidine mg range from about 1mg to 15mg per serving, meaning some retail doses sit within hailing distance of the trial range and others sit 8-12× above it, with zero trial evidence that the higher end does more.
