The honest answer

~100 mg/day is the dose behind most of the positive stress and sleep-latency signal, generally of branded PharmaGABA. 300 mg/day is the higher end tested specifically for insomnia (Byun 2018, the best-designed sleep RCT here). Retail capsules commonly sell 500–750 mg of synthetic GABA — 5 to 7.5x the trial-relevant dose — and no trial has tested whether that higher dose does more, because none actually used it for these outcomes.

The dose most of the positive human evidence uses

When researchers wanted to see whether GABA affects mood, stress reactivity, or sleep onset, most of them landed on roughly 100 mg — the dose in Yoto 2012's mental-stress EEG/mood trial and Yamatsu 2015's sleep-latency study (a 5.3-minute reduction, the modest, honest anchor number for this dose). Abdou 2006's two small relaxation studies used a similar dose class. Almost all of these trials used PharmaGABA, the branded fermented ingredient — not the synthetic GABA most retail capsules contain. And most of them (Abdou 2006, Yoto 2012, Yamatsu 2015) were funded by or authored by Pharma Foods International, the maker of PharmaGABA — a real conflict of interest to weigh against these positive results. The one trial that tested a meaningfully higher dose specifically for diagnosed insomnia symptoms, Byun 2018, used 300 mg/day of fermented-rice-germ GABA for 4 weeks with polysomnography confirmation — the best-designed trial in this evidence base, and still nowhere near the 500–750 mg most retail capsules sell.

  • Don't assume a 500–750 mg synthetic capsule is a stronger version of the same evidence. It's 5 to 7.5x the dose that was actually tested, at a form (synthetic, not PharmaGABA) that wasn't the material in the positive trials either.
  • Take it once daily, roughly 30–60 minutes before the effect you want (before a stressor, or before bed) — the closest match to how the trials dosed it.
  • On blood-pressure medication or sedatives? Check with a clinician or pharmacist first — GABA has shown a transient, moderate blood-pressure-lowering effect in some trials.
  • Pregnant or breastfeeding? No safety data exists for GABA in pregnancy or lactation (USP found none) — treat that as an absence-of-data caution, not a specific harm, and check with a clinician.
